  1. How can you increase the expressivity of the gene? Is there anyway to add the gene to the DNA repair pathway pathway?

    1. We are unsure under what conditions our gene is more highly expressed, but further experimentation could help us determine this. I also do not know of any way that we would be able to add our gene to the DNA damage response pathway.

  2. How would the gene knockout show that the gene is involved in conjugation? Basically, what would you expect as results?

    1. Looking at our gene’s expression profile, we see that it is up regulated during conjugation. This tells us that it is a part of the conjugation process, and by knocking it out we could view the impacts the absence of our gene has on conjugation. Possibly clueing us into its function during this process.

  3. Is it possible for this gene to be a part of the dna repair pathway and your experiment didn’t not catch it?

    1. I do not believe this could be the case. Since our controls yielded expected results, our results showing low expression levels of our gene are likely accurate.

    1. We exposed our T. therm cells to hydroxyurea, which creates DNA breaks by limiting the number of nucleotides available for replicating DNA.
