    2. Hi Eunvy, thanks for your question. Contamination itself adds a layer of variability in whatever results it presents itself in, which makes it a hindrance almost always. In light of out primer validation experiment (I’m assuming is where the question comes from), because of our negative and positive controls produced expected results, we could deduce the identity of the contamination. If we couldn’t do that, then our experiment would not be viable and would have to be redone.

    1. Hi Daniel. Thanks for your question! Knowing that Upec is down regulated can help us create new hypotheses of what Upec is in T.thermophila, and run different, unique experiments to explain why it shows similar expression during conjugation to other known DNA repair proteins. Additionally, if identifying DNA repair proteins is the sole purpose of another person’s research, they know they can rule out Upec for the time being and focus on a different unknown gene (lest new information be found about Upec in DNA repair).

  2. What would the fact that Upec downregulated during DNA damage indicate in terms the purpose of Upec? Would it indicate that it is involved in DNA repair at all or if it is used in other cell processes or cycles like cell death?

    1. Thanks for your question, Sarah! The down regulation does not specify anything in specific about its function and purpose in DNA damage repair. Knowing what we know so far can help us either 1.) learn more about why Upec is related to the DDR response, directly and indirectly, or 2.) redirect us to study other unknown proteins that do function.

    1. Hi Katelin! Thanks for your question. In this experiment, we used hydroxyurea (HU) to induce damage that generalized to all cells and produces certain damage. While I don’t know in specific, reagents that could target a specific cell type could be used for comparison to what we got with HU. Additionally, we could induce damage under various conditions (high temperature, pH, et cetera) to see if other stressors impact how DNA repair proteins are recruited.
