  1. If Setk is not involved in DNA damage repair, what would you guess that it is involved in? Do you know anything about the domain that might clue you in?

    1. The vWFA domain is involved with extracellular plasma proteins that aid in coagulation, haemostasis, basal cell membrane formation and intracellular proteins that have the function of transcription, DNA repair, and membrane transport. This leads us to believe that Setk may be a more involved with the transcription or membrane transport function of the domain.

  2. Since Setk doesn’t repair damaged DNA, what do you think the two peaks that indicated DNA damage repair in the conjugation phase might indicate?

    1. We discovered that upregulation during conjugation was no significant enough to indicate direct DNA damage repair. This suggests indirect involvement in DNA damage repair.

  3. Do you have an idea of what the source of contamination could have been? And if it wasn’t contaminated, what do oyu expect the results would have looked like?

    1. We determined that the RPPO in the primer validation PCR was contaminated due to the band present in the negative control with water, and the identical bands in RPPO with cDNA, gDNA, and water. For gDNA and BioBudz, we determined human error. Without the contamination and error, we expected a band for gDNA and BioBudz at 282 bp, a band for gDNA and RPPO near 800 bp, and no band with RPPO and water.

    2. We determined that RPPO was contaminated due to incorrect band with gDNA and band presence with the negative control water. The missing band with BioBudz and gDNA was caused by human error. Without contamination or error, we expected a band at 282bp with gDNA and BioBudz, a band around 800bp with RPPO and gDNA, and no band with RPPO and water.

  4. I liked ur poster a lot! What kind of experiments would you be interested in running, if any, to get a better idea of what the function might be?

    1. I would want to redo this experiment first to confirm these findings without the factor of contamination. If that confirms that Setk is not directly involved in DNA damage repair, I would want to see if it is indirectly involved because its peaks in conjugation indicate some form of involvement.
