Nice job on your presentation. You were very clear throughout. What is a gene expression profile and why is it significant to your hypothesis? Why would contamination cause one of your bands not to show up?
A gene expression profile displays the conjugation of a gene throughout the hours the cell is being observed, it is significant because Hyplrr’s expression profile is how we formed our hypothesis and is the reason we thought to experiment on our gene. RAD51 is a gene involved in DNA repair and shows a similar gene expression profile to Hyplrr, this was the origin of our hypothesis. Contamination of our cDNA with our gDNA would cause our primers to not anneal to the intended cDNA therefore annealing to nothing and expressing nothing.

We factored in a few things when we were determining our primer sequence; we designed our primers considering size, melting temperature, GC content, the likelihood of homodimer formation, and the chance of binding to a different part of our template.

Great work! How did you determine what sequence to use to make the primers for your experiment?
We factored in a few things when we were determining our primer sequence; we designed our primers considering size, melting temperature, GC content, the likelihood of homodimer formation, and the chance of binding to a different part of our template.
Nice job! How did you trigger the DNA damage in the cells when you were measuring the difference in protein expression?
We induced DNA damage by adding hydroxeurea to our treated group of DNA!
