    1. One of my TAs suggested that RAD51 is usually expressed at a higher level in cells that DNA damage has been induced in. This might suggest that DNA damage might have been induced at a smaller scale than anticipated or the higher expression of Setk might not be related to the DNA damage being induced at all.

  1. I like your poster; it is very organized. You also spoke clearly as well. My question is, what exactly does “kneeling” mean in the context of your presentation? 🙂

    1. In the context of PCR and PCR primers, a primer will anneal or bind to a template piece of DNA. This allows the DNA polymerase to replicate and copy the section of the gene that the primers specify. Our first experiment which is represented by the first figure was to make sure that our primers that we created annealed or binded to Setk correctly.

  2. Why do you believe your data conflicts with most? Do you believe it is most likely due to a mistake.? If SetK is not involved in DNA repair what is?

    1. The other groups in the Cell Biology Lab that were also studying the same gene found that the expression remained the same when DNA damage was induced which is different than upregulation our data showed. It definitely could be due to a mistake or just by chance in the data. Lots of the genes that we studied in this class had not be studied before and no one knows the purpose of the protein that Setk codes for. Setk could have many different uses during conjugation of Tetrahymena.

    1. We wanted to minimize the likelihood that the primers would bind to other genes in the Tetrahymena genome and we wanted to minimize the likelihood of the primers binding to each other to form dimers. We used Primer3 Plus as a software to do that for us and we chose the primer set that looked like it met those criteria the best.
