Running a restriction digest using specific enzymes would cut the DNA of the phage isolated and then after, the gel would show the fragment sizes. Using the website phage enzyme tools, we can compare the specific fragment sizes of other known phages, to the one we isolated and if they match its most likely in that specific cluster.
We didnt know exactly which phage we isolated however, most phages that were isolated with M.smeg in my lab did end up being temperate phages. Overall though, it was a mystery whether it would be temperate or not
