    1. Hi, I was thinking of doing a western blot simply to have another method of measuring gene expression to ensure if there is decreased expression of our gene of interest or not.

  1. Great presentation! I appreciated your introduction where you provided more information about the DNA damage and the use of RFC1 gene. Also, I’m excited to hear that your primers worked! Regarding your future directions, if you were to conduct a protein localization experiment, what information would you predict that experiment would give you? Would that information encourage you to choose a different gene to study?

    1. Wow great question! If I were to do a protein localization experiment, I would predict the protein would wither not be expressed or it would be located on the DNA during DNA damage. This could possibly help decide whether the protein is present or not during DNA damage. Considering our results, I would like to study a different gene that hopefully increases in expression after DNA damage.

  2. Could you explain a bit more on how you made the primer? Also how will the western blot test help you understand the gene expression?

    1. Yes of course! We used a software called Primer3Plus. First we set parameters that would give us a primer size product of 100-600. Next we compared the GC% and e value. Then we came across a primer pair that fit our criteria and ordered them.
