With Rfc1 being involved in DNA repair pathway we can see illnesses such as cancer that have many rounds of division. As cells divide, many instances of DNA damage often occur which cancer cells have good mechanisms to combat breaks. Being able to understand genes involved in DNA damage will help research of DNA damage repair such as cancer cells which could in turn lead to increasing the longevity of the human lifespan.
With the two separate genomes replication as well as the way the gene cuts up its entire genome with each replication causes the single celled organism to excel in DNA damage repair pathway. Rfc1 was looked at as a hypothesized gene to be involved in the DNA damage repair pathway.
Primer design was looking at different sequences on both sides of the gene of interest. We evaluated different sequences on different criteria such as if secondary structures like hairpins were expected to form, the GC composition, as well as their band lengths. Making sure to evaluate different sequences based on our criteria was very important in finding a primer set that was both useful for evaluation as well as could anneal to the genome.
