Hello! we used computer programs such as Benchling and Primer3Plus in order to design primers that would specifically amplify and anneal to the cDNA of our Rsp1 gene during a polymerase chain reaction, or PCR.
Overall, we measure the changes in gene expression to see if RSP1 was involved in the DNA Damage/repair pathway. Specifically, it can associate with RdRPs to facilitate the development of sRNAs that are needed for RNAi.
