    1. To clarify, our primer validation was more so inconclusive rather than unsuccessful. We had an issue with the PCR tubes and when they ran in the thermocycler there was some noticeable evaporation that compromised our results, that likely why we didn’t see any bands for some of our samples. Without proper control groups we can’t validate the primer and decided to use a different primer for the next experiment. If time permitted, we would have repeated this experiment with better quality samples.

Unfortunately, I wish I had a more exciting response but the RFC subunit 1 was assigned to me by the course instructor so I didn’t have a choice in which subunit was involved in this research. However, it would be interesting to repeat this experimentation with other subunits as well!

  2. Hi Allison, great question! cDNA is codingDNA or complimentaryDNA, meaning it is the part of the genome that only includes coding regions, exons and not introns. Whereas gDNA, genomic DNA, is the entire organisms genome including various non-coding regions. gDNA does not vary, however, cDNA is more specific to the point it is isolated from in terms of tissue type, cell type, or conditions. In this experiment, it is important to see that our gene is present in the cDNA during cell conditions that require proteins involved in DNA replication and damage.
