    1. If we were to repeat the experiment we do believe that our positive controls could work if we better performed the experiment. Because the RPPO did not increase in expression as it was supposed to, that meant that all of our experimental results could not be used/trusted.

    1. We were led to that conclusion because within our positive control, an increase of expression and a brighter band would be present if induced. Because the band stayed exactly the same, we were able to conclude that the only explanation would be that damage was not induced because no other variable factors were present.

  1. What were you expecting if the experiment did work as far as in the gels? (Aside from the positive control)

We were expecting our gene specific results to better match the consensus gels meaning that the band would have increased in brightness for each of the primer lanes, not just for the positive control. We were expecting this because our gene is involved in DNA damage repair so its band should have increased in brightness.

    I was wondering if you had any potential explanations for the positive control results.

    1. The explanation for the positive control results is that we believe we did not correctly induce DNA damage. If we had induced DNA damage, the brightness would have increased in the second lane of the positive control showing that the gene expression was also increased.
