  1. Could you describe the process of selecting your primers? Were there qualitative aspects of T. thermophilia that caused you to select those specific primers?

    1. The primers were based on the base pairs present in the gene itself, and verifying that those sequences don’t show up too frequently in other regions of the genome. This would minimize amplification of any undesired sequences.

  2. You mentioned in future directions that you would do a western blot to measure the levels of the
    Twi2 PIWI protein within the cell. What exactly is this and why is knowing these levels important?

  3. With Twi2 being involved in the repair of DNA damage how will use continue with research about this gene? what does this genes involvement in repair say about how Twi2 can be used or altered to possibly treat cancer?

  4. Do you think the Twi2 protein would be expressed more, less or just as much if the DNA were exposed to say UV light opposed to the hydroxyurea?

    1. Honestly I’m not sure but I can say that the damage induced by hydroxyurea is much faster than if it was exposed to UV. It would have to be exposed under a UV light for much longer than it takes to dose the cells with hydroxyurea.

    2. I’m honestly not sure, however I would speculate that the UV light would require more exposure time than the hydroxyurea.
