It was actually determined previously that the genes involved in sRNAs are essential for proper DNA damage, as they saw accumulation. TWI8 is one of these biogenesis proteins so this does indicate there is a possibility it plays a direct role in DNA damage repair.
What does the literature say about Twi8 expression in single strand breaks? Are there any other experiments on Twi8 expression that you would need to do to test this?
I’m not very knowledgable of the changes in expression of TWI8 due to single stranded breaks. From our lab the general consensus was that TWI8 increased in expression due to double stranded breaks, which may indicate TWI8 plays a role in DNA damage repair. As for testing single stranded breaks we would first need to determine an agent that does this efficiently, and whether or not it may be diluted enough to cause a low enough number of breaks where it would likely not affect the gene sequence itself. Following this with very similar procedures as what was done with double stranded breaks.

An example of this could be apoptosis where a cell is unable to undergo proper DNA damage repair, and how this may relate to cancer in having cells that are immune to this pathway and can accumulate damage with no regulation on their life cycle.
You mentioned that the gel contained errors that skewed your results. Do you have any idea what could’ve caused these errors for the sake of future experimenters?
Yeah! We are pretty sure that our gel apparatus was on a slant so the way it solidified caused one of the corners with wells to be at a higher level so the wells were smaller due to this and we weren’t able to load the right amount of sample.
