We came to this conclusion as Narcan works as an antibiotic at high concentrations. These high concentrations are not safe for the human body generally speaking.
A macrophage is a cell that surrounds and kills microorganisms, removes dead cells, and stimulates the action of other immune system cells. Narcan is commonly used to prevent opioid overdosing. It does so by attaching to to the opioid receptor in the macrophage. Thus, in general, Narcan would be a good candidate for an antibiotic since it benefits the macrophage.
If we were to continue testing at the 300-400 microliter dilution we would indeed keep everything else the same. This is to ensure all our controls are the same and have reasonable data to compare it to. Testing at a smaller dilution between these concentrations will allow us to track down when Salmonella begins to grow. This is in order to figure out when our drug will stop the inhibition of Salmonella Typhimurium.
During our trials, we left the first row and column empty we believed there was a manufacturing error in these plates. In previous weeks, when we tested plates, data from many plates were concluded to be inadequate. Data from the first column and first row were different than the rest of the wells tested. Therefore, we tried to avoid any possible mistakes to ensure we get accurate data to analyze.
Generally speaking, we can achieve 10 μM of any given compound in our blood. We don’t want to test higher than this because if it only worked at a dose that is higher than what we can get into someone’s system, it wouldn’t be safe to test in humans.
