Hi Elena, restriction enzymes recognize a specific part of the DNA called palindromic sequence, and can cut that specific, recognized sequence. The six enzymes here were already chosen for us, and I am assuming that the reason for choosing these particular enzymes is that they have the capability of cutting the desired sequence but not the capability to cut elsewhere such as the promoter region or the origin of replication because the then the DNA cannot be replicated.
Hi Grace, Restriction enzyme result shows whether or not your phage genomes is similar to other mycobacteriophages. Phage genomes that are similar will have a similar restriction cut sites, and you can see that on your gel electrophoresis. The gel will show the number of cuts that can be compared to other mycobacteriophages and if they are similar, it means that they have the same cluster, but it is only a prediction, must be confirmed by doing genome sequencing.
Hi Jackson, the temperate phage’ DNA is incorporated into the host cell DNA, but this doesn’t mean that it doesn’t go through lytic life cycle. It goes through both life cycles, and the turbid plaques shows that because not all the bacterial particles are destroyed, so you see a cloudy plaque around the clear circle. Now, if it only goes through lytic life cycle, it immediately can lyse the host bacteria, and that’s why you can only see a clear plaque. Hope this makes sense.

Our result shows that our phage is a lytic phage, and it has a high capability of lysing the host cells. The quality and the quantity of the DNA is pretty good. These are some of the factors that make a phage a good candidate for phage therapy.
