Great job to both of you! Were you guys ever able to determine where your DNA became damaged during the duration of your lab? This would be super beneficial for anyone to know who wants to repeat your experiment(s)! 🙂
We weren’t able to determine where the DNA became damaged since we went straight into restriction digest and we didn’t have time to redo our DNA purification.
I think the fact that we were only able to do DNA purification once affected our results because we didn’t know if our purification had worked and based off of our gel results the purification didn’t work as well as we would have liked.
We ran our gel for about 20-30 minutes and this was determined by our lab manual. We stopped running the gel once the bromophenol blue was about 80% down the gel.
There have been some studies where temperate phages have been genetically engineered, so I think our phage will be able to be used in future phage research.
