When my phage is characterized, it can be sent to Actinobacteriophage Database. Once there, it has the potential to be used in phage therapy depending on the bacteria that needs to be infected.
To isolate my phage, I had to use a soil sample and make a lysate out of it to then test the sample if it has any potential phages. To do this, I plated the lysate and looked for any plaques in the agar. Since the plate did result in plaques, I was able to pick a plaque and perform three rounds of purification to isolate the phage to one species. The purification process is lengthy, and consisted of me diluting my phage sample (using phage buffer) many times in each round of purification to see which dilution resulted in isolated plaques. After the third round of purification we were able to find an isolated plaque, and use it to make the high titer lysate.

I would say just being able to find a phage from my soil samples because I was concerned I wouldn’t be able to find one.
