  1. Why do you think only the cDNA sample showed a positive result with the control primer in the first gel (Fig. 3)?

We wondered this as well, and as a lab we all came to the conclusion that either the gDNA was too large to exit the well during the time that the gel was run for (the gDNA was ~2000 bp) or the gDNA was degraded because everyone in the lab had the same results of no gDNA bands being shown in the positive control.

  2. What other iterations of the experiment indicated that Hypec was more expressed after being induced with DNA damage, and why do you think that is?

    1. The consensus gel (a validated result of Hypec’s DNA damage results) as well as others in our lab who studied Hypec indicated that Hypec was more expressed. These results indicate that Hypec is involved in the DNA Damage Response pathway.

  3. What other iterations of the experiment indicated that Hypec was more expressed after being induced with DNA damage, and why do you think these results were different?

The results were the same as ours, as ours also indicated increased expression of Hypec; However, we were unable to verify our result because our positive control that would indicate that the same amount of cDNA was put into each sample was degraded, so we could not say for sure in our results that the brighter bar in the gel was caused by increased expression or an error in loading the sample.
